Vav1 structure unveils its mechanism of action
Rapley and colleagues have solved the crystal structure of Vav1, a guanine nucleotide exchange factor, bound to its substrate – Rac1. The structure reveals that the C1 domain does not contact the GTPase, as previously thought, but forms a unit that stabilizes the active conformation of the DH domain, promoting its exchange activity. This work provides the basis for understanding the relative promiscuity of Vav1 and for resolving the many discrepancies in the field.

DasR links nutrient stress and antibiotic production
Soil-dwelling Streptomyces produce secondary metabolites, including antiobiotics. In this study, Rigali et al, report that GlcNAc is a crucial signaling molecule that ultimately results in the inhibition of the repressor DasR, therefore triggering development and antibiotic production. This work unveils the first complete signaling cascade from nutrient sensing to morphological development and antibiotic biosynthesis.

RalA maintains Jak/Stat signaling in egg chambers
Polar cells and outer border cells play an important role in establishing egg patterning in Drosophila. In this study, Ghiglione et al, identify RalA as a crucial factor in polar cell differentiation and survival during oogenesis, and show that RalA is required cell autonomously for expression of polar cell specific markers and for secretion of the Jak/Stat ligand Upd. These results identify RalA as a novel regulator of patterning and migration in Drosophila.
